A recent study published in the journal Nutrients has revealed that omega-3 fatty acids, commonly found in fish oil, may play a significant role in reducing insulin resistance and improving glucose tolerance in non-obese rats. This research sheds light on the potential benefits of omega-3 supplementation for individuals who are not obese but still suffer from conditions akin to type 2 diabetes, characterized by elevated blood sugar levels due to impaired insulin function.
The study focused on Goto-Kakizaki (GK) rats, which serve as a model for non-obese type 2 diabetes. Researchers administered fish oil supplements containing 540 mg of eicosapentaenoic acid (EPA) and 100 mg of docosahexaenoic acid (DHA) per kilogram of body weight, three times a week over an eight-week period. The results indicated a notable reduction in insulin resistance, alongside improvements in blood glucose levels and various lipid profiles, including total cholesterol and triglycerides.
According to the authors, while omega-3 fatty acids have been previously recommended for patients with cardiovascular issues and type 2 diabetes, their effects on insulin resistance in non-obese individuals have not been thoroughly explored.
Rui Curi, a prominent researcher involved in the study, explained that the findings suggest omega-3 supplementation can shift the inflammatory response in these rats from a pro-inflammatory state to an anti-inflammatory one. This modulation in the immune response is crucial, as chronic low-level inflammation is known to interfere with insulin signaling pathways.
The research team observed changes in lymphocytes, which are white blood cells essential for the immune response. In non-obese rats with insulin resistance, these lymphocytes were found to produce higher levels of pro-inflammatory cytokines, a pattern similar to that seen in obese individuals with diabetes. The primary objective of the study was to determine if fish oil supplementation could reverse the inflammatory changes previously noted in these non-obese rat models.
Renata Gorjão, a co-author of the study, emphasized the importance of understanding the relationship between inflammation and insulin resistance, even in the absence of obesity. This knowledge could pave the way for new treatment strategies for the approximately 10% to 20% of diabetes patients worldwide who are non-obese.
The broader implications of this research highlight that while obesity is a significant risk factor for diabetes, it is not the sole contributor. Genetic factors may also play a role in the development of insulin resistance among non-obese individuals. Previous studies have indicated that systemic inflammation exists in non-obese rats with insulin resistance, a condition that differs from the inflammation typically associated with increased adipose (fat) tissue in obesity.
Interestingly, the study also pointed to an early breakdown of anti-inflammatory pathways in young GK rats, suggesting that the immune system's ability to regulate inflammation may be compromised from a young age in those predisposed to insulin resistance. The researchers found that fish oil supplementation effectively reversed the inflammatory profile, enhancing the presence of regulatory T-cells, which help inhibit pro-inflammatory responses.
While the results are promising, the researchers caution that further studies are necessary to confirm these findings and to determine the optimal dosages and types of omega-3 fatty acids beneficial for human patients. The ongoing investigation aims to deepen the understanding of insulin resistance mechanisms in non-obese populations and explore potential therapeutic interventions.
