Cervical cancer ranks among the most prevalent gynecological cancers globally and remains a significant cause of cancer-related mortality among women. Despite advancements in early detection through screening methods, treatment options for advanced or recurrent cervical cancer are still quite limited. The five-year survival rate for patients diagnosed with late-stage cervical cancer remains below 20%, highlighting the critical need for more effective therapeutic interventions.
This type of cancer is primarily associated with high-risk human papillomavirus (HPV) infections, although various other factors contribute to tumor development, including immune system changes, abnormal cellular survival signals, and increased angiogenesis that fosters tumor growth. Current treatment protocols, including surgery, radiation, and chemotherapy, tend to be more effective in the initial stages of cancer but offer reduced efficacy once the disease has progressed.
Recent research conducted by a collaborative team from Jiangxi Provincial Cancer Hospital and Case Western Reserve University has identified the potential of andrographolide, a compound derived from the medicinal plant Andrographis paniculata, in combating cervical cancer. Known for its anti-inflammatory and anti-cancer properties, andrographolide has displayed promise in various cancer studies, yet its specific effects on cervical cancer had not been thoroughly investigated prior to this research.
Published in the journal Biomolecules and Biomedicine, this study employed a unique hybrid tumor model known as CAM-PDX, which integrates a patient-derived xenograft (PDX) system with a chicken egg chorioallantoic membrane (CAM) platform. This innovative model facilitates the growth of human tumor tissue within a biologically relevant and cost-effective environment, making it ideal for preclinical drug testing.
Through experiments utilizing tumor tissue from a patient suffering from cervical squamous cell carcinoma, the researchers discovered that andrographolide significantly inhibited tumor growth and reduced angiogenesis, or new blood vessel formation. These anti-cancer effects were notably enhanced when andrographolide was administered alongside cisplatin, a commonly used chemotherapy agent.
Microscopic examinations revealed that tumors treated with andrographolide exhibited a less organized structure and a reduced number of blood vessels compared to untreated samples. The treatment also resulted in lowered levels of key proteins associated with cell survival and angiogenesis, including Ki67, BCL-2, and ERG. This suggests that andrographolide functions by not only inhibiting cell proliferation but also restricting the tumor's blood supply.
The findings of this study underscore the potential of the CAM-PDX model as a rapid and economical approach for assessing new oncological therapies. Furthermore, the results position andrographolide as a promising candidate for future drug development, particularly in combination with existing treatment modalities.
Additional research is warranted to further explore how andrographolide interacts with the immune system and whether it can augment immunotherapy strategies. Nonetheless, these discoveries represent a significant advancement toward developing more effective treatment options for individuals diagnosed with advanced cervical cancer.
