The team found that mitochondria within CA2 neurons are diverse in structure and function, depending on their location. Mitochondria located at the farthest ends of the dendrites, where synaptic connections are made, are particularly specialized and heavily rely on MCU for proper functioning.
By genetically modifying mice to delete the MCU gene in CA2 neurons, researchers noted a disruption in plasticity at the outermost synapses, while connections closer to the neuron's cell body remained unaffected. This implies that the diversity of mitochondrial function is not merely incidental but a crucial aspect that allows different parts of the neuron to operate distinctly.
The implications of this research extend to various neurological disorders, including Alzheimer's disease and autism spectrum disorder. Mitochondrial dysfunction has been increasingly linked to these conditions, impacting the energy supply necessary for sustaining synapses and, consequently, cognitive functions.
In Alzheimer's, the outermost synapses are often the first to experience decline, suggesting that disruptions in MCU function within CA2 neurons might contribute to this vulnerability.
